Huan is not just any wolfhound, he was enhanced by the Valar. These are divine beings created by Eru Ilúvatar. He received immortality, no longer needed to sleep, and was allowed to speak 3 times before he died (you can learn more about him in the lore article about him).
After he rescued the elf maiden Lúthien, the daughter of the then king Thingol, they were attacked on their journey by werewolves of Sauron. Together they were able to defeat them. Sauron himself then confronted the two in battle as the greatest werewolf that ever existed. He did this due to a prophecy that stated that Huan would die by the hand of a werewolf.
Sauron attacked Lúthien, but she blinded him and Huan attacked. Sauron could not defeat the wolfhound and fled by transforming into a vampire, basically a bat creature. Ultimately, Huan died in battle against Carcharoth, a werewolf bred by Morgoth.
